Pengaruh BA dan Thidizuron Terhadap Inisiasi dan Multiplikasi Tunas Tapak Dara

Abstract


Tapak dara (Catharanthus rosea L.) is One of ornamental plants having medicinal Propeeties .To intiate crop improvement Through tissue culture it is necessary to Provide aseptic plant materials. Plant Multiplication through in vitro culture has been conductued at plant Generic Resource and Breeding Laboratory in Research Institute for Spice and Medicinal Crops Bogor. Single node explants from Cimanggu experimental garden were used the explants were planted on Murashige and Skoog (MS) medium sipplemented with 30 g/l agar. The experiment was divided in two stages. The first stage is preliminary experiment and the second stage is shoot multiplication. For the first stage single node stems were cultured on MS Medium suplemented with BA (0.1, 0.3 and 0.5 mg/l) to find right concentration of BA For shoot intiation. The result from the first Stage is used as explant for the second stage For the second stage BA concentration was Increased to become 0.5, 1.0, 2.0 and 3.0 mg l And supplement with 0.1 mg/l thidiazuron. The experiment was design randomized Complete (CRD) with 10 replications.The Result showed that the treatment of BA 0.5 Mg/l was the best medium for shoot intitation With green, fresh and vigorous clture. Treatment with BA 0.5 mg/l +thidiazuron 0.1 mg/l gave the fastest growth with average number of shoots, leaves and height werw 9.29, 36 and 3.63 cm respectively. The best Performance of cultures with green, fresh Leaves and vigorous stem were obtained from The same medium
